Аннотация
BACKGROUND: Recently, we developed a novel sandwich enzyme-linked immunosorbent assay (ELISA) for measuring serum agalactosyl-immunoglobulin G (agal-IgG) using the 42B1 monoclonal antibody. Serum agal-IgG is significantly elevated in patients with chronic inflammation, including chronic hepatitis. Some patients with adult congenital heart disease (ACHD) also have hepatic congestion. Hence, we explored agal-IgG as a potential biomarker in patients with ACHD.
METHODS: A total of 87 patients with ACHD who underwent cardiac catheterization were retrospectively analyzed. Clinical and catheter data were collected at the Osaka University Hospital between 2019 and 2024. Their serum agal-IgG was determined using a sandwich ELISA.
RESULTS: The level of log-transformed agal-IgG was positively correlated with inferior vena cava pressure (IVCP), and IVCP was a significant independent variable for log-transformed agal-IgG. In terms of biochemical data, serum B-type natriuretic peptide (BNP), gamma-glutamyl transferase (γ-GT), total bilirubin (T-Bil), alkaline phosphatase (ALP), and Mac-2 binding protein glycosylation isomer (M2BPGi) were positively correlated with log-transformed agal-IgG and negatively correlated with high-density lipoprotein cholesterol (HDL-C), and sex (female vs male). Also, multiple regression analysis for IVCP as the objective variable and log-transformed agal-IgG, T-Bil, M2BPGi, γ-GT, ALP, BNP, HDL-C, and sex as explanatory variables showed that log-transformed agal-IgG had a beta coefficient of 3.786 (p = 0.030), which meant a significant independent variable for IVCP.
CONCLUSION: In patients with ACHD, serum agal-IgG levels are significantly associated with increased IVCP.
