Аннотация
Uterine fibroids and obesity are comorbid conditions with similar pathogenetic mechanisms. The aim of this study was to analyze the association of GWAS-significant uterine fibroid loci with the risk of obesity. DNA genotyping of 115 obese and 218 non-obese patients for 17 SNPs was performed by real-time PCR. An association was established between rs58415480 SYNE1 (effect G allele; OR = 0.61; p = 0.019) and rs11031731 THEM7P/WT1 (effect A allele; OR = 0.63; p = 0.04) with a reduced risk of obesity. Bioinformatic analysis showed that the alleles modulate the binding sites of transcription factors that cause inhibition of adipogenesis, regulation of glucose and lipid metabolism, as well as the retinoic acid and Notch signaling pathways, which indicates the pleiotropic role of these loci and their involvement in both the proliferative and metabolic pathways.
