Аннотация

Chidamide, a novel inhibitor that selectively targets histone deacetylase (HDAC)1/2/3/10, has been approved in China for treating peripheral T-cell lymphoma and breast cancer. Previous studies have indicated the involvement of certain histone deacetylases in renal fibrosis (RF) progression. However, the potential therapeutic effect of chidamide in RF management remains unclear. In the current study, the effects and potential mechanisms by which chidamide alleviates RF were investigated. The influence of chidamide on the expression of epithelial-mesenchymal transition (EMT)-related indicators was assessed in renal tubular epithelial cells induced by transforming growth factor-β1. Moreover, whether chidamide delays RF progression was explored in C57BL/6J mice via unilateral ureter obstruction (model 1) and high-dose folic acid injection (model 2). RNA sequencing (RNA-seq) analysis was employed to predict the pathways through which chidamide alleviates RF. In fibrotic human kidney 2 cells and primary renal tubular epithelial cells, chidamide reduces the expression of EMT-related proteins. In the RF mouse models, chidamide significantly reduces pathological damage in fibrotic renal tissues, reduces serum creatinine and blood urea nitrogen levels, alleviates tubular injury, reduces collagen deposition, and decreases the expression of RF markers. RNA-seq and biochemical analyses further suggest that chidamide elicits an antifibrotic effect through the phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT) signaling pathway. Collectively, these results suggest that chidamide may inhibit RF progression through the PI3K/AKT signaling pathway.
