The expression and significance of CXCL13 and CXCR5 in the tumor microenvironment of primary central nervous system diffuse large B-cell lymphoma.

BACKGROUND: PCNS-DLBCL (Primary central nervous system diffuse large B cell lymphoma) is a rare extranodal non-Hodgkin lymphoma. It is limited to the brain, eyes, spinal cord, and leptomeninges. In PCNS-DLBCL, we explore the correlation between the CXCL13/CXCR5 axis and the Tumor microenvironment (TME)and the ratio of peripheral blood lymphocyte subpopulations. We also explore the possible mechanisms by which the CXCL13/CXCR5 axis regulates the TME of PCNS-DLBCL, and the impact on its prognosis.
METHODS: We performed Multiplex immunohistochemical(mIHC) and immunohistochemical (IHC) on paraffin-embedded tumor tissues from 25 patients with PCNS-DLBCL. The localization and expression levels of CXCL13 and CXCR5 in PCNS-DLBCL tumor tissues were analyzed, and correlated with pathological features, survival and other clinical parameters. We analyze the correlation between CXCL13/CXCR5 and the infiltration of immune cells (T cells, macrophages), the expression of immune checkpoints (PD-L1, CD39).
RESULTS: CONCLUSIONS: The CXCL13/CXCR5 axis may exert a bidirectional regulatory effect of anti-tumor immunity and tumor immune evasion through multiple mechanisms, such as influencing the infiltration of immune cells, the expression of immune checkpoints, and the immune function of the peripheral blood, which ultimately affects the therapeutic efficacy and prognosis of PCNS-DLBCL.
