Аннотация
An improved method of cannulation of the bile duct and duodenum in rats has been developed to study the kinetics of medicinal compounds in the bile. The proposed double cannulation method ensures reliable fixation and accurate placement of the catheters, prevents their twisting and loss, and allows for a comprehensive study of the processes of substance absorption and elimination. The bile was collected from animals without anesthesia, the circulation of bile acids between the liver and intestine was preserved. This method has been tested in a study of the kinetics of 2,6-diisobornyl-4-methylphenol in rat bile over 24 h after oral administration (25 mg/kg in 0.5 ml of linseed oil). The pharmacokinetic profile of the substance has been shown to be a plateau with an indistinct maximum (216.87 ± 74.39 ng/ml, after 8 h), and its slow excretion with bile is observed.
