New Insights into the Pathogenesis of Psoriatic Arthritis.

In the current paradigm, psoriatic arthritis (PsA) pathogenesis is generated and sustained by combined innate and immune disease mechanisms dominated by the IL-23/IL-17 axis. The current challenge is to define tissue-specific endotypes and associated biomarkers with the goal to develop a precision medicine-based approach. In this article, we discuss the current pathogenetic landscape in PsA and outline the path forward to define tissue-specific endotypes. A more complete understanding of the cellular and molecular heterogeneity in specific target tissues and the development of models that recapitulate human disease will accelerate the rationale development of tissue domain-guided personalized therapies in PsA.
