Аннотация

The recent study by de Siqueira Santos et al. (J Neurochem. 2025; 169 (8): e70194) represents a significant contribution by identifying Cerebral Dopamine Neurotrophic Factor (CDNF) as a potent inducer of neuritogenesis in dorsal root ganglion (DRG) neurons via the PI3K/Akt pathway. While we commend the authors on their robust in vitro findings, which position CDNF as a promising therapeutic agent for peripheral neuropathies, our letter provides a critical analysis to contextualize and extend these discoveries. We posit that the mechanistic narrative, while compelling, remains incomplete. The exclusive reliance on pharmacological inhibition to establish PI3K dependency warrants validation through isoform-specific genetic approaches, and the potential for critical signaling crosstalk with pathways such as Ras/MAPK is largely unexamined. Furthermore, the study's reductionist model, utilizing isolated DRG neurons, overlooks the integral role of satellite glial cells in modulating neuronal responses in vivo. The most substantial gap illuminated is the enigmatic identity of the putative CDNF receptor, a fundamental knowledge gap that must be bridged to fully understand the specificity and regulation of CDNF signaling. We conclude that while the study by de Siqueira Santos et al. provides a foundational pillar, the translation of these findings will necessitate a more comprehensive mapping of the signaling network, identification of the receptor, and rigorous validation in pathologically complex in vivo models that recapitulate the challenging microenvironment of nerve injury.
