Effect of protease inhibitors from Siparuna brasiliensis leaves on experimental autoimmune encephalomyelitis.
OBJETIVE: Protease inhibitors have emerged as promising candidates for modulating autoimmune diseases. This study evaluates the pharmacological potential of protease inhibitors extracted from leaves of Siparuna brasiliensis in an experimental autoimmune encephalomyelitis model, a preclinical model of multiple sclerosis.
METHODS: Female C57BL/6 mice were immunized with the MOG35-55 peptide to induce EAE. The mice were then divided into groups and treated with either PBS or Siparuna brasiliensis protease inhibitors at 500 mg/kg or 1000 mg/kg via oral gavage. The study assessed clinical scores, central nervous system histopathology, and cytokine levels (IL-12p70, IFN-γ, IL-6, IL-17, and IL-10). Additionally, the identity of the active protein components was confirmed using SDS-PAGE and NMR analysis.
RESULTS: demonstrated that protein extract treatment significantly reduced clinical disease scores, neuroinflammation, and brain and spinal cord demyelination. Additionally, the inhibitors modulated inflammatory cytokine levels, promoting a beneficial immune response. SDS-PAGE and NMR analysis identified active protein components in the extract with molecular weights of approximately 12 and 15 kDa, consistent with Bowman-Birk-type protease inhibitors.
CONCLUSION: These findings highlight the neuroprotective and immunomodulatory effects of Siparuna brasiliensis protease inhibitors, suggesting their potential as therapeutic agents for autoimmune diseases.
