Аннотация

BACKGROUND: Chronic hyperglycemia can cause oxidative stress damage to the liver, which is one of the main complications of diabetes. Cysteine (Cys), acting as a key thiol involved in maintaining the redox balance of the liver, plays an important role in the pathogenesis of diabetic liver injury. There is currently a lack of means for real-time monitoring of the dynamic changes of Cys in the liver. Therefore, the development of related technologies is crucial for revealing the potential pathological mechanisms of diabetes.

RESULTS: Here, we developed a near-infrared (NIR) fluorescent probe that detects Cys with high selectivity via an ester-bond cleavage mechanism. The probe showed a clear fluorescence turn-on response to Cys, with its emission peak at 735 nm allowing for deep-tissue imaging. Background autofluorescence was largely avoided, and the detection limit reached 0.2 μM. When applied to hyperglycemic hepatocytes, the probe uncovered a clear relationship between glucose concentration and intracellular Cys levels, revealing a pronounced depletion of Cys under high-glucose conditions. In a diabetic mouse model, in vivo imaging further showed a marked reduction in hepatic Cys signals compared to healthy controls, confirming the oxidative stress-associated Cys decline in the liver. This work provides a practical molecular tool for dissecting the role of Cys in diabetes-related liver injury.

SIGNIFICANCE: The Cys-responsive NIR fluorescent probe enables effective tracking of changes in Cys levels in hyperglycemic hepatocytes and diabetic liver tissues. It not only advances our understanding of the pathological mechanisms underlying oxidative stress-induced liver injury but also provides a reliable experimental basis for further investigation into the pathogenesis of diabetes and its associated pathological processes.
