Аннотация

The extraction of indomethacin from biological samples was carried out using a new sorbent that employs (hydrated vanadium pentoxide (V2O5·1·6H2O)) in a dispersive solid phase extraction process. Indomethacin, a commonly used nonsteroidal anti-inflammatory drug, is frequently present in complex biological matrices, making its accurate extraction crucial for pharmacokinetic studies and therapeutic monitoring. The proposed method in this study used of the distinctive properties of V2O5·1·6H2O, which has high selectivity and adsorption capacity for indomethacin, enabling effective separation from interfering substances. Optimization of crucial parameters such as sorbent amount, extraction time, and elution conditions were conducted to maximize extraction recovery of the analyte. The method exhibited significant enhancement in extraction efficiency and clean up indomethacin in plasma. Furthermore, the extracted analyte underwent analysis using high performance liquid chromatography coupled with a tandem mass spectrometry, achieving limits of detection and quantification that comply with regulatory standards. This study underscored the potential of V2O5·1·6H2O as a cost-effective and efficient material for the proposed extraction method, contributing to improved analytical methodology for the detection and quantification of pharmaceuticals in plasma assessments. Validation results indicated a wide linear range (5-150 ng mL-1), acceptable coefficient of determination (0.995), and low limits of detection (0.2 ng mL-1), and quantification (0.7 ng mL-1), as well as good precision (relative standard deviations ≤ 6.0% for intra-day (n = 7) and inter- (n = 5) precisions). These findings demonstrated the high potential of the proposed method for screening indomethacin in human plasma samples.
