Tradipitant for Motion Sickness: Expanding the Therapeutic Landscape of NK₁ Receptor Antagonists.

OBJECTIVE

To review the efficacy, safety, and tolerability of tradipitant for the prevention of vomiting induced by motion.

DATA SOURCES: A literature search was conducted through PubMed using the following terms: tradipitant, motion sickness, Nereus, and neurokinin-1 (NK-1) receptor antagonist.

STUDY SELECTION AND DATA EXTRACTION: Articles describing the pharmacology, pharmacokinetics, efficacy, safety, and/or tolerability of tradipitant were included in this review.

DATA SYNTHESIS: Tradipitant is a NK-1 receptor antagonist shown to reduce vomiting due to motion. Clinical trials have demonstrated improvements in the occurrence of vomiting as well as multiple episodes of vomiting secondary to various sea conditions. Common adverse effects associated with this agent include fatigue, somnolence, and headache when taken as a single-dose treatment.

CONCLUSIONS

Scopolamine and first-generation antihistamines are commonly used to prevent and treat motion sickness; however, their use is limited by their significant adverse effect profiles. Tradipitant, the newest agent approved for motion sickness, provides clinicians with an additional treatment option for this common condition.
