Аннотация

BACKGROUND: Preeclampsia (PE) is a high-risk complication that endangers both maternal and fetal health. Astragalus species exhibit potent antioxidant effects, which have been explored in the management of diverse diseases.

STUDY OBJECTIVE: This research aimed to explore the possible therapeutic benefits of Astragalus using a rat model of PE induced by NG-nitro-L-arginine methyl ester (L-NAME).

METHODS: Key parameters assessed in pregnant rats included 24-h urinary protein, blood pressure, placental morphology (diameter and weight), and fetal outcomes (pup length, weight, and malformation rate). Serum placental growth factor (PLGF) and soluble fms-like tyrosine kinase-1 (sFlt-1) were analyzed via enzyme-linked immunosorbent assay (ELISA). Oxidative stress markers-malondialdehyde (MDA) and glutathione peroxidase (GSH-Px)-were measured in placental tissue and serum, while catalase (CAT) and superoxide dismutase (SOD) were evaluated in serum. Placental expression of nuclear factor erythroid 2-related factor 2 (Nrf2), Kelch-like ECH-associated protein-1 (Keap1), and NAD(P)H quinone oxidoreductase 1 (NQO1) was examined using real-time quantitative PCR (RT-qPCR) and Western blot.

RESULTS: The findings demonstrated that Astragalus treatment alleviated maternal pathological manifestations and ameliorated adverse fetal outcomes in the PE model. These protective effects were partly mediated by its antioxidant properties, which involved activation of the Nrf2/Keap1/NQO1 pathway.

CONCLUSION: The results indicate that Astragalus holds promise as a therapeutic candidate for novel PE treatment strategies.
