BACKGROUND: Aerobic exercise and motor skill training independently influence hippocampal plasticity; however, their interaction when motor learning occurs under conditions of exercise-induced exhaustion remains incompletely understood.

OBJECTIVE: This study examined whether combining progressive aerobic exercise performed to exhaustion with rotarod motor training differentially regulates long-term potentiation (LTP) and BDNF/TrkB signaling in the dentate gyrus of male Wistar rats.

METHODS: Male Wistar rats were assigned to control, aerobic training (ET), rotarod training (RT), combined aerobic-rotarod (ERT), or sequential rotarod-aerobic-rotarod (RERT) groups. Training was conducted three times per week for eight weeks with progressive increases in intensity and duration. Aerobic sessions were performed to exhaustion. In combined protocols, rotarod training was administered before and/or after exhaustive aerobic exercise. Field excitatory postsynaptic potential (EPSP) slope and population spike (PS) amplitude were recorded in the dentate gyrus to assess LTP. Hippocampal BDNF and TrkB protein levels were quantified using ELISA.

RESULTS: A significant group-by-time interaction was observed for EPSP slope (p < 0.05), with ET exhibiting the greatest potentiation. In contrast, PS amplitude demonstrated time-dependent modulation without significant between-group differences. BDNF levels were highest in ET, whereas repeated rotarod exposure was associated with lower BDNF expression relative to ET. TrkB levels were elevated in ET and ERT compared with controls (p < 0.05).

CONCLUSION: These findings indicate that combining progressive aerobic exercise with motor skill training modulates hippocampal plasticity in a context-dependent manner rather than producing linear additive effects. The dissociation between dendritic (EPSP) and somatic (PS) responses suggests differential regulation of synaptic efficacy and neuronal output when motor learning occurs under exercise-induced exhaustion.
