What's already known about this topic? ◦. Pathogenic PITX1 variants, including deletions and missense mutations, give rise to a syndromic phenotype characterized by clubfoot, tibial hemimelia, and preaxial polydactyly. ◦. To date, no previously reported case of PITX1 variant or deletion has featured upper‐limb involvement. What does this study add? ◦. The PITX1 p.Ala123Thr variant is associated with a markedly distinct clinical presentation involving anomalies of both upper and lower limbs. ◦. Our cases expand both the phenotypic and genotypic spectrum of the PITX1‐related limb malformations.
