BACKGROUND: Angelicae Sinensis Radix (AS), a commonly used herb in traditional Chinese medicine, demonstrates antidepressant effects potentially linked to erythrocyte homeostasis regulation. However, the precise mechanism by which AS modulates erythrocyte homeostasis to achieve its antidepressant efficacy remains to be elucidated.
PURPOSE: This study investigated the mechanism by which AS regulates erythrocyte homeostasis in a chronic unpredictable mild stress (CUMS) rat model of depression.
METHODS: Iron metabolism and hematopoietic function in rats subjected to CUMS were assessed using an enzyme-linked immunosorbent assay (ELISA), while erythrocyte metabolic features were characterized using lipidomics. To ensure robustness and biological relevance of the findings, an advanced and rigorous multi-parameter screening strategy was introduced and applied for identifying differential metabolites. This integrated model incorporates VIP, |log2(FC)|, -ln(p-value), and |p(corr)| values, and significantly enhances the reproducibility and precision of metabolite identification, which is particularly advantageous for deciphering complex systems such as herbal pharmacology. Western blotting was employed to validate the results screened by the novel strategy, followed by correlation analysis to elucidate the relationship between sphingolipid metabolism and erythrocyte homeostasis.
RESULTS: Significant changes were observed in iron metabolism-related indexes, erythrocyte antioxidant-related indexes and erythropoietic function indexes of the rats underwent the modeling process, which were mitigated by the administration of AS. Significant changes in erythrocyte lipid metabolites and key enzymes of sphingolipid metabolism disrupted the homeostasis of erythrocytes. The administration of AS significantly regressed this situation.
CONCLUSIONS: AS may regulate red blood cell homeostasis and exert antidepressant effects through the sphingolipid metabolism pathway.
