From network prediction to experimental validation: Multi-Target mechanisms of Artemisia absinthium L. Essential Oil (AAEO) against depression.
Аннотация
ETHNOPHARMACOLOGICAL RELEVANCE: Artemisia absinthium L. (wormwood) has a long-standing tradition in European and Central Asian folk medicine for its neuroregulatory properties, historically used to calm emotions, alleviate nervousness, insomnia, and dizziness. Despite this ethnomedicinal precedent suggesting potential benefits for mood disorders, the systematic investigation of its antidepressant material basis and mechanisms remains unexplored.
AIM OF THE STUDY: This study aimed to elucidate the multi-target mechanisms underlying the antidepressant effects of A. absinthium L. essential oil (AAEO), bridging its traditional neurocalming uses with modern pharmacological validation.
MATERIALS AND
METHODS
GC-MS characterized AAEO composition. Network pharmacology predicted active compounds, core targets, and pathways, validated by molecular docking and dynamics simulations. In vivo efficacy was assessed in chronic unpredictable mild stress (CUMS)-induced depressive mice (behavioral tests: sucrose preference, open field, forced swimming). In vitro, corticosterone (CORT)-injured PC12 cells evaluated AAEO's cytoprotection (viability, LDH, apoptosis, mitochondrial potential). Core target necessity was functionally validated via siRNA knockdown and Western blotting.
RESULTS
GC-MS identified 98 compounds in AAEO, predominantly terpenoids (40.8 %) and esters (22.4 %). Network pharmacology prioritized alpha-thujone, linalyl phenylacetate, and mansonone C as core compounds. Molecular docking confirmed their strong binding affinities (-8.80 to -5.34 kcal/mol) to GABAergic (GABRA2, GABRB3) and serotonergic (SLC6A4, MAOB) targets, further stabilized in 200-ns molecular dynamics simulations. In vivo, AAEO (100-300 mg/kg) significantly ameliorated CUMS-induced depressive behaviors (restored sucrose preference, locomotor activity, reduced despair). In vitro, AAEO (5-20 μg/mL) protected PC12 cells against CORT-induced apoptosis and mitochondrial dysfunction, concurrently upregulating GABRB3 and SLC6A4 expression. Critically, siRNA-mediated knockdown of GABRB3 or SLC6A4 significantly abolished AAEO's cytoprotective effects.
CONCLUSION
AAEO exerts significant antidepressant effects through a multi-target mechanism involving synergistic actions of its key bioactive compounds (alpha-thujone, linalyl phenylacetate, mansonone C) on GABAergic (via GABRB3) and serotonergic (via SLC6A4) systems. This study provides mechanistic validation of A. absinthium L.'s traditional neurocalming uses, positioning AAEO as a promising multi-target phytotherapeutic candidate for depression.
