Аннотация
Prolonged or excessive inflammatory response can result in various ailments. Angelica sinensis (AS) is a classical medicinal and edible plant. Wine-processed AS (WAS) is widely applied to treat arthritis and other inflammatory diseases. Our study discovered that dichloromethane fractions of WAS (WASD) exhibited superior anti-inflammatory activities compared to dichloromethane fractions of AS (ASD). However, the mechanism remains indistinct. This study aimed to elucidate the potential mechanism by an integrated method conjoined chemical analysis, network prediction and experimental verification. UHPLC-MS identified 32 and 27 constituents from ASD and WASD, respectively. Additionally, the contents of 11 indicative and shared compounds derived from ASD and WASD were quantified, among which 5 compounds showed significant differences. Network pharmacology predicted 47 hub genes and identified PI3K/AKT, JAK-STAT and NF-κB pathways associated with the inflammation. Experimentally, the protein expressions related to Akt/NF-κB and STATs pathways influenced by WASD were higher compared to ASD in LPS-induced RAW264.7 cells. Furthermore, WASD exhibited stronger inhibitory effect on xylene-induced auricular edema than ASD. Summarily, WASD exceeds ASD in terms of anti-inflammatory effects in vivo and vitro. The study provides some evidence for the application of ASD and WASD in ameliorating inflammation.
