Аннотация

Human milk-derived extracellular vesicles (HMEV) are promising supplements for infant formula, as they contribute to the health benefits of breast milk. Efficient isolation of pure, intact extracellular vesicles (EV) remains challenging due to the complex milk matrix. This study compared common HMEV isolation methods-polyethylene glycol (PEG)-based precipitation, ultracentrifugation (70,000 × g, 100,000 × g, and 100,000 × g, repeated 3 times), and 0.45-µm filtration-to identify protocols optimizing yield, integrity, and purity. Before isolation, samples underwent preliminary purification to remove cells, fat, and proteins. Isolated vesicles were characterized according to MISEV2023 guidelines using nanoparticle tracking analysis for size and concentration, flow cytometry, and confocal microscopy for membrane integrity, and western blotting for EV markers (CD9, TSG101, HSP70) and the absence of contaminants (Calnexin). Polyethylene glycol precipitation yielded the highest particle concentrations (>1 × 1012 particles/mL) but slightly reduced vesicle integrity. Ultracentrifugation produced lower yields but maintained morphology and marker expression, especially at 100,000 × g. Filtration resulted in high counts but poor specificity and irregular morphology. Within the constraints of a single-donor design and based on the analytical endpoints assessed here, PEG precipitation (particularly when combined with deproteinization) and ultracentrifugation provided the most favorable trade-offs between yield, purity, and structural preservation, and these patterns should be confirmed in larger multidonor studies to evaluate their generalizability across donors. These findings also emphasize the critical role of preliminary purification in achieving high-quality HMEV preparations. To minimize biological variability and focus on method-dependent effects, all isolations were deliberately performed on aliquots of the same donor milk sample, allowing the observed differences to be attributed primarily to the isolation protocols rather than to the starting material.
