Аннотация
The allyl-arene scaffold plays a crucial role in modern drug discovery and development owing to its remarkable structural versatility, broad bioactivity, and extensive pharmacological potential. Its significance is further strengthened by recent synthetic advancements, such as allyl-allyl cross-coupling reactions, which enhance both its chemical diversity and medicinal applicability. The structural adaptability of allyl-arenes allows for the design of molecules with tailored biological properties, making them valuable frameworks in medicinal chemistry. Among various derivatives, hydroxychavicol, a naturally occurring allylarene compound isolated from Piper betle leaves, has emerged as a bioactive molecule with exceptional therapeutic promise. It exhibits diverse pharmacological effects, including anticancer, antibacterial, anti-inflammatory, antiviral, antioxidant, anticholinesterase, and platelet aggregation inhibitory activities. The molecular architecture of hydroxychavicol enables it to modulate multiple signaling pathways, such as NF-κB, COX-1/2, MAPK, JNK, AKT/mTOR, and RANKL, which are central to inflammation, apoptosis, and cancer progression. Importantly, hydroxychavicol shows cytotoxicity against a range of cancers, including chronic myeloid leukemia, breast, pancreatic, prostate, colorectal, glioblastoma, liver, oral, and skin cancers. Its bioactivity is largely attributed to its metabolic conversion into reactive electrophilic intermediates, such as ortho-quinone and para-quinone methide, which can induce DNA damage and confer antibacterial properties. The presence of two hydroxyl groups and an allylarene moiety enhances its reactivity, stability, and binding capacity, attracting strong interest from synthetic and medicinal chemists. Overall, hydroxychavicol represents a multifunctional and structurally versatile lead compound with great potential in therapeutic development.
