Аннотация

BACKGROUND

Xylene, a traditional clearing agent in histopathology, has raised concerns due to its toxicity and environmental impact. Safer, eco-friendly alternatives such as coconut oil (CO) and groundnut oil (GO) are being explored, but their efficacy in histopathology remains underexplored.

AIM: This study aimed to evaluate and compare CO and GO with xylene as clearing agents, focusing on section cutting, cellular architecture, staining quality (cytoplasmic and nuclear), cell morphology, and staining clarity.

MATERIALS AND

METHODS

Sixty tissue specimens were divided into three groups: Group X, Group CO, and Group GO. Histopathological protocols were followed, and tissue sections were stained with H and E. The tissue quality was evaluated on five parameters, and statistical analysis was performed using SPSS.

RESULTS

All groups performed similarly in section cutting and cellular architecture. Xylene and CO showed superior staining in both nuclear and cytoplasmic areas, while GO had lower scores, particularly in cytoplasmic staining. Both CO and xylene maintained good cell morphology and staining clarity, whereas GO showed inconsistencies. Statistical analysis revealed significant differences (P < 0.001) between xylene and the oils, particularly in staining quality.

RESULTS

CO showed comparable results to xylene in most parameters, while GO showed more variability, particularly in staining quality. CO proved to be a more reliable, eco-friendly alternative to xylene in histopathology, while GO showed promise but with some inconsistencies.

CONCLUSION

CO is a viable, cost-effective, and eco-friendly alternative to xylene in histopathology, with similar performance across key parameters. GO, although effective, showed some variability in staining, limiting its potential as a consistent substitute for xylene.
