Аннотация
BACKGROUND: Male sexual dysfunction, particularly erectile dysfunction and reduced libido, affects a significant portion of the global male population. While synthetic drugs like phosphodiesterase-5 inhibitors provide short-term relief, they often cause adverse effects. Natural alternatives, especially phytochemicals like coumarins, have shown promise as safer, multi-target solutions to erectile dysfunction.
OBJECTIVES: This review aims to explore the aphrodisiac potential of coumarins by examining their chemical nature, natural sources, historical and cultural significance, mechanisms of action, preclinical and clinical evidence, and future therapeutic prospects for male sexual health.
METHODS: An extensive literature review was conducted across ethnopharmacological records, biomedical databases, and clinical trial repositories. Relevant studies were retrieved from Scopus, Google Scholar, and Web of Science between 2015 and 2025 using keywords related to coumarins, aphrodisiac activity, sexual dysfunction, and male reproductive health. The review focused on studies evaluating the hormonal, neurological, vascular, and reproductive effects of coumarin compounds through in vitro assays, animal models, and limited human trials.
RESULTS: Coumarins influence male sexual health through several mechanisms: enhancing testosterone levels, modulating nitric oxide and cyclic GMP signaling, promoting vasodilation, stimulating neurotransmitters like oxytocin and dopamine, and improving sperm quality and motility. Preclinical studies show improvements in sexual behavior, erectile response, and hormone regulation. A clinical trial on Angelica sylvestris-derived coumarins demonstrated positive effects on libido and erectile function with minimal side effects. However, bioavailability issues and limited human data remain challenges.
CONCLUSIONS: Coumarins show potential as natural agents to enhance male sexual function. Their multi-target actions warrant further development via standardized dosing, safety evaluation, innovative delivery systems, and larger clinical trials to translate preclinical findings into clinical practice.
