BACKGROUND: MEK inhibitors have been widely used in clinical trials and have achieved favorable anti-tumor growth and metastasis effects, greatly improving patients' progression-free survival (PFS). Therefore, mining genes with similar function to MEK may provide more molecular targets with clinical value for anti-PCa metastasis.
METHODS: RNA-seq data were obtained from the TCGA and GEO databases for bioinformatics analysis. genes functionally similar to MEK were screened in combination with the GeneCards database and identified the key gene set. Multivariate Cox regression model identified the independent risk factors. GSCA database clarified the correlation between risk factors and epithelial-mesenchymal transition (EMT). HPA database and tissue microarray validated the clinical risk features of the target genes. In vitro experiments verified the role of a target gene in LNCaP cell function, EMT phenotype and PCa biomarkers. CIBERSORT algorithm assessed the effect of the target gene on immune infiltration in PCa microenvironment.
RESULTS: Three functional genes (HRAS, MAPK11 and MAP3K11) were highly expressed in tumor tissues with poor PFS. This gene set was a risk factor for PCa patients' PFS, in which MAPK11 and MAP3K11 were independent risk factors. MAPK11 and MAP3K11 expression was positively correlated with EMT activation. Additionally, MAP3K11 had a higher proportion of expression and was correlated with high-risk clinical features. MAP3K11 silencing significantly inhibited LNCaP cell proliferation, migration, invasion and EMT phenotype, and significantly down-regulated PSA and Prostein expression, while its overexpression had the opposite effect. Low MAP3K11 expression promoted plasma cell and regulatory T cell infiltration.
CONCLUSION: Targeted inhibition of MAP3K11 resists PCa metastasis by limiting EMT onset and mobilizes immune infiltration.
