OBJECTIVE: The growth of endothelial cells and their resistance to inflammation seriously affect the repair and regeneration of blood vessels, the integrity of vascular structure, and the cellular immune response function. At present, many materials are capable of regulating the growth and inflammatory response of vascular endothelial cells. However, their regulatory effects are not ideal and still need to be optimized.
METHODS: This study utilizes layer by layer assembly technology to assemble Chitosan (CS) with Zn-0.8Li alloy, functionalized the surface of the alloy, and forms a CS thin film on the surface of Zn-0.8Li to synthesize CS-Zn-0.8Li material. Moreover, human umbilical vein endothelial cells are used as experimental subjects. CS-Zn-0.8Li and the extraction solution of the control material are added to the cell culture medium, and after 24 h of cultivation, the cell activity and proliferation ability are detected by MTT assay. The levels of pro-inflammatory and anti-inflammatory factors such as TNF - α and IL-6 are measured using an inflammatory factor detection kit, and the expression levels of inflammatory response signaling pathway molecules are analyzed.
RESULTS: At a concentration of 20% in the leaching solution, the activity of cells can reach 91.2%, which is higher than the 83.7% of Zn-0.8Li. In addition, the proliferation ability of cells in the new material can reached 66.7%, which is 6.9% higher than that of Zn-0.8Li. The regulatory effect of the new material on inflammatory response is analyzed. It is found that after 24 h of cultivation, the content of anti-inflammatory factors in the 20% concentration of the new material culture dish is greater than 75%, and the content of pro-inflammatory factors is less than 16%, indicating a relatively low content. In both Zn-0.8Li and the new material culture dishes, the expression level of inflammatory signaling pathway molecules in the new material culture dish is less than 13%.
CONCLUSION: The above results indicate that the proposed Zn-0.8Li alloy fused with CS can promote cell proliferation ability and inhibit inflammation related factors, thereby regulating the growth of vascular endothelial cells and inflammatory response.
