RORα as a context-dependent transcriptional stabilizer in the nervous system.
Аннотация
RORα has been implicated in neural development, circadian regulation, neuroimmune signaling, and neuroprotection, though these functions are typically discussed in isolation. In this review, we propose that these diverse roles can be more coherently understood by viewing RORα as a regulator that helps sustain transcriptional programs in a context-dependent manner. Rather than functioning as a switch for a single pathway, RORα helps sustain adaptable gene-expression programs. We begin by outlining the molecular basis of RORα function. We then examine how RORα contributes to neural development, inflammatory homeostasis, circadian regulation, and cellular resilience under metabolic, ischemic, and degenerative stress. Overall, this perspective offers a more integrated framework for interpreting the diverse functions of RORα in the nervous system and may help guide future research into neural resilience and disease vulnerability. By positioning RORα as a regulatory hub within the nuclear receptor network, this framework also provides a distinctive and translationally relevant perspective for understanding the pathogenesis of complex neurological disorders and developing therapies that combine circadian modulation with neuroprotection.
