Аннотация

Tooth extraction sockets represent a particularly demanding wound environment, characterized by spatial confinement, high moisture, continuous mechanical disturbance, and exposure to a dense oral microbiota. Effective healing requires localized therapeutic strategies that can be stably retained at the wound site while delivering stage-specific interventions in a temporally coordinated manner. In this study, we developed an injectable Pluronic F127 diacrylate (PF127DA)/methacrylated hyaluronic acid (HAMA) hybrid hydrogel as a locally retained and clinically practical therapeutic platform for potential extraction socket management. The hydrogel exhibits favorable injectability, rapid photo-induced gelation within 3 s, robust tissue stability, tissue adhesion, and controllable degradation, enabling stable localization within extraction sockets. As a multifunctional carrier, it supports the temporally coordinated release of amoxicillin, ropivacaine, and fibroblast growth factor-2 (FGF-2), providing early antibacterial protection, sustained local analgesia, and prolonged pro-regenerative factor availability in accordance with wound healing progression. In vitro studies demonstrated that the hydrogel system exhibited effective antibacterial activity and a sustained ropivacaine release profile. Importantly, the hydrogel system also showed attenuation of excessive pro-inflammatory activation together with a modest shift toward repair-associated macrophage features. Overall, this study presents a material-mediated strategy that integrates robust tissue stability, temporally coordinated multi-drug delivery, and inflammatory attenuation within a single injectable hydrogel platform, offering a potential approach for the localized management of extraction sockets and other clinically challenging wounds.
