Аннотация
Epidermal growth factor receptor (EGFR) is the most frequent alteration in isocitrate dehydrogenase - wild-type glioblastoma and has been considered a promising therapeutic target. Despite extensive investigation, EGFR-targeted therapies have not improved overall survival because of limited central nervous system drug delivery, tumor heterogeneity, adaptive resistance, and an evolving tumor microenvironment. This review summarizes the molecular biology of EGFR, clinical experience with EGFR-targeted therapies, mechanisms underlying therapeutic resistance, and emerging strategies that may improve future clinical outcomes.
