STATEMENT OF PROBLEM: Silver nanoparticle (AgNP)-modified denture base materials can reduce biofilm-induced oral inflammatory lesions such as denture stomatitis because of the release of bioactive silver ions. However, whether the release of silver ions is sustained when exposed to human saliva and their effect on the molecular structure of the denture base resin are unclear.

PURPOSE: The purpose of this in vitro study was to assess the influence of salivary media on the sustained release of silver ions from a modified-denture base resin and to determine their effect on the molecular structure of the denture material.

MATERIAL AND METHODS: Acrylic resin disks (ADs) containing 0.0% (control), 1%, 0.5%, and 0.05% AgNPs (n=10) were exposed to human saliva and artificial saliva for 6 months. and silver ion release was assessed at various time points (day 0, 15, 30, 45, 90, 120, 150, and 180 days) using inductively coupled plasma-mass spectroscopy. Prior to the silver release and Raman spectroscopy study, 2 commonly used salivary substitutes (wet mouth and biotene oral rinse) were assessed for their susceptibility to Candida albicans biofilm formation on ADs, with saline used as a control. Data were analyzed using 1-way ANOVA to evaluate the relationship between the salivary substitutes and fungal biofilm (α=.05). The salivary substitute demonstrating the least biofilm formation was used for silver release and Raman analysis. Raman spectrometry was performed using a 514-nm argon laser to qualitatively assess the molecular vibrational characteristics of the modified resin on day 0, 3, and 6 months and compared with that of the control specimens.

RESULTS: A time- and concentration-dependent silver release was seen from the modified denture base resin up to 90 days in human saliva, while artificial saliva (wet mouth) demonstrated minimal release (0.2 mg/L). Additionally, no changes in the structural characteristics of the modified acrylic resin were noticed in Raman analysis across the 6-month study in natural saliva, even at the highest concentration tested (AgNP 1%).

CONCLUSIONS: The type of salivary medium significantly affected silver ion release from AgNP-modified PMMA denture base resin. PMMA resin incorporated with ultrasmall AgNPs (<10 nm) at 1% exhibited sustained silver ion release in human saliva for extended periods without compositional modification, warranting further in vitro and in vivo investigations.
