PURPOSE: To investigate whether monochromatic ambient light modulates lens-induced myopia (LIM) progression in guinea pigs and whether these effects are associated with changes in Wnt/β-catenin signaling in the retina-choroid complex.

METHODS: Forty 3-week-old male pigmented guinea pigs were randomly assigned to four groups (n = 10/group): control, broadband white light with LIM (BW-LIM), middle-wavelength green light with LIM (MW-LIM), and short-wavelength blue light with LIM (SW-LIM). Bilateral LIM was induced using -10.0 D lenses for 4 weeks. Axial length and refractive error were measured weekly. Ocular structural changes were evaluated by fundus photography, optical coherence tomography, histology, and TUNEL staining. Expression of Wnt2b, Fzd5, and β-catenin in the retina-choroid complex was assessed by RT-PCR and Western blot.

RESULTS: After 4 weeks, compared with the control group, the BW-LIM group showed an axial elongation of 0.25 ± 0.02 mm and a myopic shift of -4.9 ± 0.3 D (both P < 0.0001). Relative to the BW-LIM group, the MW-LIM group showed significantly less axial elongation (0.16 ± 0.02 mm vs. 0.25 ± 0.02 mm, P = 0.025) and a smaller myopic shift (-3.75 ± 0.3 D vs. -4.9 ± 0.3 D, P = 0.005), whereas the SW-LIM group did not differ significantly from the BW-LIM group. Reduced axial elongation under middle-wavelength light was mainly attributable to less vitreous chamber deepening (difference vs. BW-LIM: 0.10 ± 0.03 mm, P = 0.03). All LIM groups showed choroidal and scleral thinning, which was attenuated in the MW-LIM group but not in the SW-LIM group. No overt retinal structural damage or increase in apoptosis was detected under either monochromatic lighting condition. At the transcriptional level, Wnt2b, Fzd5, and β-catenin were significantly upregulated in the BW-LIM group versus controls. In the MW-LIM group, expression levels were lower than those in the BW-LIM group, whereas the SW-LIM group showed no significant difference from the BW-LIM group. Protein expression showed a similar pattern.

CONCLUSIONS: Ambient middle-wavelength green light attenuated vitreous chamber deepening and slowed LIM progression in guinea pigs and was associated with downregulation of Wnt/β-catenin signaling in the retina-choroid complex. Short-wavelength light showed no significant effect compared with broadband-light LIM.
