Expediting industrial expansion is contributing to global challenges attributable to unregulated discharge of hazardous pollutants, including emerging xenobiotic compounds. To explore broad-range remediating bioresource, present investigation reports, for the first time, whole-genome sequence (WGS) of novel white-rot-fungus, Trametes flavida WTFP2, an unexplored Polyporales with unique genes for tolerance to heavy-metals (HMs) and xenobiotic degradation. WTFP2 exhibited remarkable in-vitro degradation of 8 synthetic dyes and 8 HMs, making it versatile remedial bio-agent. Its bioremediation potential was elucidated by WGS and annotations. The de-novo assembled genome was 42.72 Mb, with 56.13 % GC content, encoding 17,654 predicted genes. Of these, 11,149 genes (63.15 %) have assigned functions. Furthermore, the study identified 368 CAZymes, and 16 biosynthetic gene clusters (BGCs) for terpenoid biosynthesis, lignocellulose and xenobiotic degradation. Genome mining revealed candidate genes for lipase, and remediation of sundry xenobiotics, representing HMs, synthetic dyes, dioxin, benzoate, toluene, polycyclic-aromatic-hydrocarbon, atrazine, phenol, and fluorobenzoate. A novel degradation pathway for enzymatic-biomineralization into TCA cycle was constructed. Comparative genomics via variant calling and annotation identified various markers and 7546 unique genes. Moreover, comparative proteomics detected 11,710 orthologs, with ∼6245 shared across genomes. Phylogenetic analysis revealed its closest relative as Trametes coccinea. Thus, T. flavida-dependent mycoremediation might be benchmark for water-pollution mitigation and sustainable-development-goals.
