Аннотация
Salt-inducible kinase 2 (SIK2) and poly (ADP-ribose) polymerase 1 (PARP-1) form an unexpected, intricate synergistic network driving tumorigenesis, therapy resistance, and immune evasion, positioning their co-inhibition as a transformative strategy to overcome fundamental limitations in current cancer therapy. Mounting mechanistic evidence underscores their synergistic interplay in perpetuating genomic instability, enhancing tumor cell survival under replicative stress, and evading immune surveillance. Critically, emerging data suggest that SIK2 and PARP-1 engage in a functionally integrated regulatory network extending beyond their roles, collectively driving tumor progression and resistance. Simultaneous targeting of these nodes may thus yield unique therapeutic advantages by overcoming the intrinsic limitations of conventional therapies. This review integrates mechanistic insights, structural biology, and drug design perspectives to establish a novel framework for developing dual SIK2/PARP-1 inhibitors, offering a promising path to redefine treatment paradigms for recalcitrant cancers.
