Аннотация

Polysaccharide copolymers Conjuates have surfaced as a versatile foundation in the development of advanced smart drug delivery systems, owing to their inherent biocompatibility, biodegradability, and capacity for chemical modification. This review brings into focus the recent advances in co-polymeric drug delivery systems based on naturally occurring polysaccharides like chitosan, alginate, dextran, hyaluronic acid, pullulan, guar gum, xanthan gum, agarose, gellan gum, and starch. Their structural malleability and functionalization capabilities are emphasized to engineer therapeutic payload stability, bioavailability, and controlled release. The synthesis and design methods of Polysaccharide copolymeric conjugates are explained, coupled with advanced drug encapsulation techniques, conjugation, and surface modification approaches towards site-specific delivery. Mechanistic understanding of targeted delivery routes receptor-mediated targeting, tumor microenvironment responsiveness TMR, and pH/enzyme-sensitive release are discussed to exemplify the specificity and efficacy of these systems in oncology. The review explores the multifunctional applications of Polysaccharide copolymeric conjugates in cancer therapy, such as chemotherapy, gene and siRNA delivery, and immunotherapy. The advances in nanomedicine, especially in nanoparticle engineering, stimuli-responsive systems, and overcoming multidrug resistance MDR, are also discussed. By integrating the recent trends and innovations in Polysaccharide -based copolymeric drug delivery systems, in this article encapsulates the potential of these biopolymers as next-generation nanocarriers in targeted cancer treatment. The challenges and future directions in clinical translation are critically evaluated to steer future research towards more effective and safe therapeutic platforms.
