Аннотация

BACKGROUND: Emerging evidence suggests that neurofilament light (NfL) can serve as a blood-based biomarker for diabetic neuropathy (DN), a severe complication that affects half of patients with type 2 diabetes mellitus (T2DM). Fiber optic particle plasmon resonance (FOPPR) technology, which integrates optical waveguides with noble metal nanoparticles, offers highly sensitive detection of target analytes. It can potentially be applied to measure NfL in blood samples.

METHODS: A FOPPR-based plasma NfL assay was developed and validated for imprecision, accuracy, limit of quantitation (LOQ), and analytical measurement range (AMR). A comparison was made between this assay and a single molecule array (Simoa) assay.

RESULTS: Plasma NfL values were measured for 25 diabetic patients with DN (T2DM with DN) and 19 diabetic patients without DN (T2DM). It was observed that the plasma NfL values were significantly elevated in the T2DM with DN group in comparison to the T2DM group.

CONCLUSIONS: A plasma NfL assay has been successfully developed based on the FOPPR technology. The assay offers a promising approach for early clinical diagnosis of DN.
