Аннотация

Bacopa monnieri (BM), a traditional Ayurvedic herb, has emerged as a promising multifunctional therapeutic plant for managing type 2 diabetes mellitus (T2DM). This comprehensive review examines the molecular, cellular, and systemic mechanisms underlying the antidiabetic effects of BM and its key phytoconstituent, bacoside A. Evidence from in vitro, in vivo, and silico models reveals that BM improves insulin sensitivity, restores β-cell functionality, suppresses hepatic gluconeogenesis, and attenuates oxidative stress and inflammation. Notably, recent in silico docking studies demonstrate a strong interaction between bacoside-A and PPAR-γ, highlighting its potential to modulate metabolic and neuroprotective pathways. In addition to metabolic regulation, emerging evidence supports the role of BM in addressing diabetes-associated neurodegeneration-a condition characterized by insulin resistance in the brain, which is linked to cognitive dysfunction and neurodegeneration. Bacoside-A and BM extract modulate AMPA-type glutamate receptors, downregulate pro-apoptotic markers like Bax, and mitigate oxidative-nitrosative stress in neuronal tissues, thereby supporting cognitive resilience in diabetic states. These neuroprotective actions position BM as a dual-targeting botanical agent for both (T2DM) and diabetes-associated neurodegeneration. Furthermore, the integration of multi-omics data, including transcriptomics and metabolomics, has illuminated BM's biosynthetic pathways and its multitargeted pharmacological profile. Network pharmacology and protein-protein interaction analyses confirm bacoside-A's regulatory roles in NOTCH signalling, insulin receptor expression, and hepatic enzyme modulation. Collectively, these findings validate BM as a viable, low-toxicity candidate for next-generation phytopharmaceutical development in the holistic management of diabetes and its complications.
