Аннотация

OBJECTIVE: The molecular mechanisms underlying polycystic ovary syndrome (PCOS), a multifactorial disorder, remain unclear. Altered serum microRNAs (miRNAs) and circRNAs (circRNAs) in PCOS can serve as biomarkers in this disease. This study was aimed at assessing the serum levels of circRNA Pum1_0014, miRNA-146a, and miRNA-141-3p, as well as their potential as biomarkers in PCOS.

PATIENTS AND METHODS: This study examined 50 patients meeting PCOS criteria and 50 apparently healthy women as controls. Serum levels of metabolic and hormonal parameters were investigated in all participants. Real-time quantitative reverse transcription polymerase chain reaction was used to assess the expression of the three studied genes.

RESULTS: The gene expression of miRNA-141-3p and circRNA Pum1_0014 was downregulated in the PCOS group vs. the control group. Additionally, miRNA-146a was significantly (p < 0.05) overexpressed in the PCOS group vs. the control group. In the PCOS group, miRNA-146a showed a highly significant negative correlation with circRNA Pum1_0014, whereas both RNAs showed highly significant correlations with FAI and SHBG. Moreover, miRNA-141-3p showed highly significant (p < 0.001) correlations with several metabolic parameters. Therefore, circRNA Pum1_0014, miRNA-146a, and miRNA-141-3p might serve as novel diagnostic tools with high sensitivity and specificity for PCOS prediction.

CONCLUSION: The study proposes use of circRNA Pum1_0014, miRNA-141-3p, and miRNA-146a as new biomarkers in PCOS, and indicates their probable roles in the hormonal and metabolic imbalances in PCOS.
