Аннотация

Polycystic ovary syndrome (PCOS) is a prevalent endocrine-metabolic disorder lacking effective treatment strategies that allow controlled and precise drug release. In this study, an oxidation-reduction responsive nanocomposite drug delivery system, termed Maltotriose-APTMS-1@CP1@SBFI-26, was constructed. In this system, compound 1 was functionalized with maltotriose-(3-aminopropyl)trimethoxysilane (Maltotriose-APTMS) and subsequently co-encapsulated with the CB2R-selective agonist SBFI-26 within the porous coordination polymer CP1 to enable controlled drug release. The introduction of a peptide-based component enhanced system integration and improved biocompatibility. Electrochemical analysis confirmed successful drug encapsulation, with a significant drop in oxidation peak current (from 0.65 to 0.38 mA) and an increase in charge transfer resistance (from ∼950 to ∼1400 Ω). Under a constant voltage of -0.6 V, the electro-responsive release of SBFI-26 reached 68.2 μg/mL within 160 min, nearly twice the release obtained via passive diffusion. In vitro experiments further demonstrated that this nanoplatform effectively restored hormone balance and significantly reduced FOXM1 gene expression in a PCOS model. This work highlights a novel electrochemical stimulus-responsive system for controlled drug delivery, offering a promising strategy for precision therapy in PCOS management.
