ETHNOPHARMACOLOGICAL RELEVANCE: Aristolochia paucinervis Pomel is a perennial Western Mediterranean plant, traditionally used to treat wounds, inflammation, snakebites, skin diseases, cancer, infections, and pain. Despite its extensive use, studies on its phytochemistry, pharmacological properties, and safety remains limited.

AIM: This study provides the first phytochemical characterisation of A. paucinervis leaf decoction and evaluates its total phenolic content, as well as its antioxidant, anti-inflammatory, and wound-healing potential, and safety profile.

MATERIALS AND METHODS: A decoction from A. paucinervis leaf was prepared according to traditional practices. Its phytochemical profile was characterized by HPLC-PDA-MSn and TLC, quantified using HPLC-PDA, and total phenolic content assessed by Folin-Ciocalteau. Antioxidant potential was assessed using in vitro chemical assays (DPPH, ABTS, and FRAP). Anti-inflammatory activity and safety were preliminary assessed in RAW 264.7 macrophages, by NO and resazurin assays, respectively. Wound-healing potential and safety were determined using the scratch wound assay in NIH/3T3 fibroblasts.

RESULTS: HPLC-PDA-MSn analysis revealed caffeoylmalic acid as the major compound, followed by an acetyl-p-coumaroyl hexoside, puerol A, protocatechuic acid hexoside, ferulic acid and isorhamnetin-O-dihexoside. The extract exhibited total phenolic content of 109.59 ± 1.39 mg GAE/g, and demonstrated antioxidant activity across the in vitro chemical assays employed (DPPH, ABTS and FRAP). The decoction evidenced a dose-dependent inhibition of NO production in macrophages (IC50 = 34.99 ± 0.16 μg/mL), without cell-damaging effects. In NIH/3T3 fibroblasts, the extract showed the highest wound closure at 10 μg/mL, whereas both lower and higher concentrations exhibited diminished effects.

CONCLUSIONS: A. paucinervis leaf decoction results suggested antioxidant, anti-inflammatory and wound-healing activities, without cell-damaging effects, supporting its widespread traditional use in the treatment of skin lesions and underscoring its potential as a future therapeutic drug.
