Stimuli-responsive vesicles in water-resistant nanofibers for molecular random access in DNA data storage.
Аннотация
DNA is an attractive medium for data storage. Enabling efficient random access is essential for the practical use of DNA-based memory. We present a strategy for random access based on the selective release of target DNA from a nanofiber mesh matrix. Messages encoded in sets of DNA sequences are encapsulated inside stimuli-responsive small unilamellar vesicles (SUVs), embedded in an electrospun water-resistant nanofiber mesh. The target sequences are selectively released using orthogonal pH-based triggers. The orthogonality of these triggers enables the preservation of the SUVs containing the non-selected DNA, which remains in the fiber mesh for subsequent retrieval. This work combines sequence-independent chemical addressing with physical compartmentalization and double-layered protection. It represents initial steps towards a PCR-independent molecular selection system for random access in water-resistant fibers with potential applications in DNA-based memory architectures.
