Аннотация
The growing convergence between precision oncology and natural-product-based therapeutics has fostered innovative strategies that integrate molecularly targeted anticancer agents with biologically active phytochemicals to enhance therapeutic efficacy and overcome treatment resistance. Capmatinib, a selective MET inhibitor approved for MET-driven non-small cell lung cancer, and curcumin, a natural polyphenol possessing antioxidant, anti-inflammatory, and anticancer activities, represent a promising therapeutic combination. This emerging paradigm highlights the need for sensitive, sustainable, and efficient analytical platforms capable of monitoring both compounds in pharmaceutical and biological matrices. Accordingly, two new spectrofluorimetric methods were created and validated, allowing for the quantification of capmatinib and the simultaneous assay of capmatinib-curcumin combinations. The first method harnessed the unique microenvironmental properties of sodium dodecyl sulfate as a monomeric denaturant to markedly enhance the intrinsic fluorescence of capmatinib in an aqueous medium, providing a linear response over 5.0-120.0 ng/mL with a detection limit of 1.42 ng/mL. The second method exploited synchronous spectrofluorimetry (Δλ = 160 nm) to resolve overlapping fluorescence signals, enabling separation-free quantification of capmatinib and curcumin across concentration ranges of 20.0-700.0 and 10.0-250.0 ng/mL, respectively. Both methods demonstrated excellent selectivity, accuracy, and precision according to ICH guidelines and were successfully applied to spiked human plasma samples.
