Аннотация
Peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α), encoded by the PPARGC1A gene, operators as a master transcriptional coactivator in energy metabolism, and is governed by an exceptionally complex promoter network. This review summarizes the multi-promoter architecture, isoform diversity, and conserved cis-regulatory elements of PPARGC1A, including critical binding sites for MEF2, ERR, CREB, FoxO, NRF and HNF-4α. This review delineates the molecular mechanisms through which nutritional signals (fasting, exercise), systemic hormones (thyroid hormone, glucocorticoids, estrogen), and cellular stress signals (NO, TNF-α, Ang II) dictate PGC-1α expression via targeted promoter modulation. The impact of epigenetic regulatory mechanisms such as DNA methylation and histone modifications are discussed. The highly tissue-specific functional profiles of PGC-1α across skeletal muscle, liver, adipose tissue, and the myocardium is summarized, highlighting its indispensable role in mediating metabolic adaptation, muscle fiber-type determination, glucose and lipid homeostasis, adaptive thermogenesis, and the preservation of mitochondrial integrity. Finally, this review outlines contemporary methodologies for promoter analysis and proposes future research trajectories. Notably, leveraging the unique metabolic and environmental adaptability of porcine models, the use of comparative genomics and targeted epigenetic strategies is advocated to decode the PPARGC1A promoter, aiming to establish a theoretical foundation for the advanced utilization of superior agricultural germplasm.
