Аннотация

Microbial cell factories have emerged as important platforms for the sustainable production of fuels, chemicals, pharmaceuticals, and other value-added bioproducts. Synthetic biology provides a systematic engineering framework for their construction and optimization through standardized biological parts, modular design, and iterative Design-Build-Test-Learn cycles. This chapter summarizes the major tools and strategies for developing microbial cell factories, beginning with the fundamental principles of synthetic biology and the characteristics of representative microbial chassis. Key genetic engineering approaches, including CRISPR-based genome editing, transcriptional and translational regulation, and modular DNA assembly, are discussed together with strategies for metabolic pathway design, enzyme engineering, flux balancing, and dynamic regulation. Genome-scale metabolic modeling, adaptive and directed evolution, and multi-omics analysis are further introduced as systems-level approaches for identifying engineering targets and improving cellular performance. In addition, emerging computational tools, machine learning, artificial intelligence, high-throughput screening, and laboratory automation are highlighted for their roles in accelerating strain development and closing the Design-Build-Test-Learn cycle. Finally, representative applications illustrate the translation of engineered microbial cell factories from laboratory-scale design to industrial biomanufacturing.
