Аннотация

Zhang et al. combine electrochemistry, metabolite identification, transcriptomics, and ribA deletion to implicate riboflavin (RF) in the response of Escherichia coli to mild heat treatment. We highlight a causal distinction that is important for interpreting the knockout experiment. Because the ΔribA strain cannot grow normally without exogenous RF, its impaired phenotype establishes riboflavin auxotrophy but does not by itself demonstrate a heat-specific protective role. The food-matrix rescue experiment further shows that extracellular RF can bypass loss of endogenous biosynthesis. In parallel, increased extracellular RF and electrochemical signals are consistent with altered redox metabolism, but do not yet establish active RF secretion or RF-mediated extracellular electron transfer. Genotype-by-temperature testing with genetic complementation, matched baseline fitness, paired intra- and extracellular RF quantification, membrane-permeability controls, and defined electron-acceptor experiments would separate essentiality from heat-specific adaptation and strengthen the translational case for ribA as a food-safety target.
