Аннотация
Nuclear receptor (NR) subfamily 4 group A member 2 (NR4A2) is an orphan NR essential for the survival of midbrain dopaminergic neurons. Featuring a unique, dynamic collapsed ligand-binding domain (LBD), NR4A2 resides in the nucleus under basal conditions but undergoes nucleocytoplasmic shuttling under pathological triggers such as oxidative stress. Small molecules modulate NR4A2 by engaging diverse domains through dynamic LBP expansion, covalent latching, surface AF-2 targeting and retinoid X receptor (RXR) heterodimerization. Gene control exerted by NR4A2 reflects the extensive binding potential of its DNA-binding domain (DBD) with monomeric NGFI-B responsive elements (NBREs) and dimeric Nur response elements (NurREs). Here, we discuss a novel gene and protein interaction network of NR4A2 that is correlated with disease association data. This network mapping advances our understanding of NR4A2 across neurodegenerative and other systemic disorders, while highlighting key translational challenges in small-molecule drug discovery.
