Аннотация

Noncoding RNAs, including long noncoding RNAs, enhancer RNAs, circular RNAs, and small nuclear and nucleolar RNAs, represent a major, yet still largely uncharacterized, regulatory layer in the eye. A subset of these molecules, chromatin-associated RNAs (caRNAs), physically interacts with DNA and nuclear protein complexes to influence genome organization, transcriptional output, and stress responses. Although caRNAs have emerged as essential regulators of chromatin topology and gene expression in diverse tissues, their identity, dynamics, and mechanisms in the retina and retinal pigment epithelium (RPE) remain elusive. Recent high-resolution genomic studies, disease transcriptomes, and mechanistic studies implicate dysregulated caRNAs in age-related macular degeneration, glaucoma, and diabetic retinopathy, yet causal chromatin-level functions have rarely been demonstrated. In this review, we summarize current understanding of ncRNA-mediated regulation in vision, highlight emerging evidence for caRNA involvement in retinal development, photoreceptor and RPE specialization, and disease-associated epigenetic remodeling, and outline major mechanistic gaps that must be addressed. We then survey the rapidly expanding CRISPR toolbox, including base and prime editors, CRISPRi/a, Cas13 knockdown and editing platforms, and RNA-recruitment and spatial repositioning systems, that now enable targeted perturbation of both caRNA genomic loci and transcripts. These technologies provide the first practical route to move beyond correlative RNA-chromatin maps toward the mechanistic dissection of how caRNAs organize regulatory hubs, modulate enhancer-promoter communication, and shape long-range genome interactions in the eye. As the catalog of pathogenic noncoding variants grows, the ability to decode and engineer caRNA networks will be central to future precision therapeutic strategies for ocular disorders.
