Jumping Translocation Breakpoint (JTB) Protein: Linking Evolution, Development, Proteoforms, and Tumor Biology.
Аннотация
Structural remodeling is increasingly recognized as an important source of functional diversity, allowing evolutionarily conserved genes to generate multiple proteoforms while retaining critical domains. The Jumping Translocation Breakpoint (JTB) gene exemplifies this paradigm. JTB encodes a highly conserved 146-amino acid transmembrane protein involved in cell cycle regulation, mitotic fidelity, apoptosis, and genomic stability. Chromosomal rearrangements, including jumping translocations (JTs), can generate isoforms and truncated variants, expanding the functional repertoire of JTB and contributing to cancer pathogenesis. Two JTB isoforms have been described: JTB1, containing a signal peptide, cysteine-rich extracellular domain, transmembrane segment, and cytoplasmic tail; and JTB2, a shorter variant lacking the signal peptide. A truncated JTB variant (JTB-T) has also been proposed, potentially producing a soluble ectodomain with altered biological activity. Structural analyses highlight conserved cysteine residues forming disulfide bonds, with the cytoplasmic C-terminal region identified as potentially pathogenic. JTB expression is broadly observed in normal tissues but is frequently dysregulated in malignancies, acting context-dependently as an oncogene or tumor suppressor. By integrating genomic, transcriptomic, and proteomic data, studies of JTB illustrate how highly conserved proteins can balance structural robustness with functional adaptability, providing insights into tumorigenesis, therapeutic response, and potential biomarker development.
