Аннотация
The genus Averrhoa includes economically and nutritionally important fruit crops like Averrhoa carambola (star fruit) and Averrhoa bilimbi (bilimbi). However, it has not been studied enough genetically, even though it is important for horticulture. For germplasm conservation, cultivar identification, and crop improvement programs, it is important to be able to accurately measure genetic diversity. This chapter elucidates the optimization and utilization of random amplified polymorphic DNA (RAPD) markers for the analysis of genetic diversity in Averrhoa species. We used a modified CTAB method to get high-quality genomic DNA from young leaf tissues. Then, we screened primers and set standard PCR conditions to get clear and repeatable amplification profiles. Out of 30 random decamer primers from different RAPD series, we chose the ones that made unique banding patterns for further study. Agarose gel electrophoresis was used to separate the amplified products, and only bands that were consistent and well-defined were scored to make sure the results were reliable. The standardized RAPD protocol described in this chapter offers a swift, economical, and reproducible method for the molecular characterization of Averrhoa germplasm. The methodology can be effectively employed for evaluating genetic variability, enhancing breeding strategies, and bolstering conservation initiatives in underutilized tropical fruit crops.
