OBJECTIVE: This study aimed to explore the effects of circ_FAM210A on the migration and invasion of NSCLC cells through the miR-670-5p/SYNE3/apoptotic pathway axis.
METHODS: The expressions of circ_FAM210A, miR-670-5p, and SYNE3 in NSCLC cells and tissues were analysed using RT‒qPCR. circ_FAM210A was characterized by Sanger sequencing, actinomycin D treatment, and RNA digestion assays. Nuclear‒cytoplasmic separation and FISH were performed to determine the distribution of circ_FAM210A. Cell models of NSCLC (A549 and H1299) were established. The effects of upregulation of circ_FAM210A expression and downregulation of miR-670-5p expression on the malignant progression of NSCLC were assessed via plate colony formation, cell scratch repair, chamber assays, apoptosis analysis, and protein band detection. The interactions between miR-670-5p and SYNE3 or circ_FAM210A were examined using dual-luciferase and RIP assays. Rescue experiments were conducted to analyse the regulatory effect of the circ_FAM210A/miR-670-5p/SYNE3 axis on NSCLC development. In vivo experiments were conducted to explore the effect of circ_FAM210A on tumours in mice.
RESULTS: In NSCLC cells and tissues, the expression of circ_FAM210A and SYNE3 decreased, whereas the expression of miR-670-5p significantly increased. Circ_FAM210A is highly stable and is mostly distributed in the cytoplasm of A549 and H1299 cells. Both the downregulation of miR-670-5p expression and the overexpression of circ_FAM210A inhibited the proliferation, migration and invasion of NSCLC cells and promoted apoptosis as well as the activity of apoptotic signals. miR-670-5p directly binds to circ_FAM210A and SYNE3. The inhibition of SYNE3 reversed the cell inhibition mediated by miR-670-5p knockdown. Furthermore, overexpression of miR-670-5p or knockdown of SYNE3 abrogated the suppressive effects of circ_FAM210A on NSCLC growth and invasion. In vivo experiments demonstrated that upregulation of circ_FAM210A expression reduced both the size and weight of tumours in mice.
CONCLUSION: Upregulation of circ_FAM210A expression can inhibit the malignant progression of NSCLC, partly through the miR-670-5p/SYNE3/apoptotic pathway axis.
