AIM: To investigate the expression, function, and mechanism of the CELSR1 protein in tubal pregnancy.
METHODS: We collected fallopian tubes from patients attending the Department of Gynecology in Jinhua Maternity and Child Health Care Hospital. CELSR1 protein expression was detected by immunohistochemistry (IHC) and western blotting (WB). Cell invasion and migration were evaluated by scratch wound healing and Transwell assays. Ciliary morphology was investigated by scanning electron microscopy.
RESULTS: CELSR1 expression was significantly higher in tubal pregnancy tissues compared to controls (P < 0.001). The knockdown of CELSR1 significantly inhibited cell invasion (P < 0.0001) and migration (P < 0.0001). No significant differences in cell proportion and the length of cilia cells were detected between the two groups (P > 0.05). CELSR1 protein expression was significantly higher in patients experiencing tubal pregnancy than in the control group.
CONCLUSIONS: Our data suggested that elevated levels of CELSR1 expression in tubal pregnancy promoted epithelial cell invasion and migration, and altered ciliary function, thus contributing to the disease process.
