Stage-specific plasma levels of IL-17 and Del-1 and their response to nonsurgical periodontal treatment.

PURPOSE: Interleukin-17 (IL-17) and developmental endothelial locus-1 (Del-1) are involved in neutrophil transendothelial migration in periodontitis. This study compared plasma IL-17 and Del-1 levels according to periodontitis severity and evaluated the effects of nonsurgical periodontal treatment on their concentrations.
METHODS: This study included 89 patients with periodontitis: 26 with stage I, 32 with stage II, and 31 with stage III disease. Blood collection and clinical periodontal examinations were performed at baseline. Patients received nonsurgical periodontal treatment and oral hygiene instructions over 2 or 4 visits. At 4-6 weeks after treatment, 48 patients, with 16 in each stage group, returned for follow-up blood collection and clinical periodontal examination. Plasma IL-17 and Del-1 levels were measured using enzyme-linked immunosorbent assays. The χ² test, one-way analysis of variance followed by Duncan's test, and the paired t-test were used. Pearson correlation coefficients were calculated to analyze correlations among IL-17, Del-1, and other variables.
RESULTS: As periodontitis severity increased, IL-17 levels increased significantly, whereas Del-1 levels decreased significantly (P<0.001). After periodontal treatment, IL-17 levels decreased in all stages, and Del-1 levels increased except in stage I. The increase in Del-1 level was significant in stage III (P<0.05).
CONCLUSIONS: IL-17 and Del-1 may be candidate biomarkers of periodontitis severity. The observed changes after nonsurgical periodontal treatment also suggest that plasma IL-17 and Del-1 levels may reflect treatment response.
