Аннотация

Coumarins, a diverse class of naturally occurring and synthetic compounds, have garnered significant attention in drug development due to their ability to modulate enzymatic activity. These molecules exhibit dual functionality, acting as both enzyme inhibitors and enzyme inducers, influencing various biochemical pathways. Their structural flexibility allows for diverse interactions with key enzymes, making them valuable in pharmacology, particularly in the treatment of cancer, cardiovascular diseases, and neurodegenerative disorders. This comprehensive review explores the intricate mechanisms through which coumarins interact with enzymes. Also, it discusses the role of these compounds in competitive, non-competitive, and mixed inhibition, where they interfere with enzymatic processes by either occupying active sites or altering enzyme conformation. Conversely, certain coumarins function as enzyme inducers, enhancing enzymatic activity through transcriptional upregulation and post-translational modifications. These properties position coumarins as promising therapeutic agents, offering both inhibitory and activating effects within the same biochemical pathways. Furthermore, this work examines how these interactions influence drug metabolism, particularly through cytochrome P450 enzymes and UDP-glucuronosyltransferases, which are critical in drug biotransformation and detoxification. The pharmacokinetic and pharmacodynamic profiles of coumarins, including their bioavailability, metabolism, and therapeutic efficacy, are also analyzed to provide insights into optimizing their clinical applications. By bridging fundamental biochemical insights with clinical potential, this in-depth review underscores the necessity of further research to refine coumarin-based therapies. Future investigations should focus on structural modifications, targeted drug delivery systems, and personalized medicine approaches to maximize their therapeutic benefits while minimizing adverse effects. As the biochemical understanding of coumarins deepens, their role in precision medicine and enzyme-targeted therapies is poised to expand, unlocking new possibilities in disease treatment and drug design.
