Applications of natural marine polysaccharide scaffold materials in periodontal tissue regeneration.

Scaffold-based periodontal tissue engineering offers a promising strategy for regenerating tissues destroyed by periodontitis, for which conventional treatments fall short. An ideal scaffold must not only possess good biocompatibility to establish a regenerative microenvironment but also address the complex pathological features of periodontitis, including persistent bacterial infection, hyperactive immune-inflammatory responses, and the demand for bone defect repair. Natural marine polysaccharides (such as alginate, carrageenan, fucoidan, chitin and its derivatives, and hyaluronic acid) exhib

AI Structural Biology in Nephrology: Powerful Scaffolds, Fragile Certainties.

Artificial intelligence (AI)-based protein structure prediction has rapidly entered nephrology, providing plausible atomic models for channels, transporters, receptors and scaffolds that previously lacked structural information. AlphaFold and RoseTTAFold now underpin mechanistic interpretations of inherited kidney diseases, including autosomal-dominant polycystic kidney disease, cystinosis and Alport syndrome, and support experimental design in cryo-electron microscopy and cryo-electron tomography. For nephrologists, this means that renal proteins no longer need to be treated as black boxes: g

Evaluation of Fungicidal Canopy Sprays for the Management of Cytospora plurivora in Western Colorado Peach Orchards.

Cytospora canker, primarily caused by Cytospora plurivora D.P. Lawr. L.A. Holland & Trouillas is one of the most destructive diseases of peach (Prunus persica [L.] Batsch), a crop with significant economic and cultural importance in Colorado. Canker pathogens including Cytospora spp. are a major driver of reduced productivity and longevity of orchards worldwide, yet management options are limited. Fungicides can be used to protect pruning wounds from infection but have primarily been tested as hand sprays which are labor intensive and therefore expensive. Air blast sprayers are currently used

Scaffolding a Low-Cost Experiential Bioreactor Module: Reducing Cognitive Load to Enhance Competency and Self-Efficacy.

Low-cost experiential modules are critical for democratizing biotechnology education, yet most undergraduates lack hands-on experience with continuous-culture bioreactor operations, which are fundamental to industrial bioprocessing. This study employed design-based research across two iterative cycles (Fall 2024, n = 8; Fall 2025, n = 11) to develop a cost-effective experiential laboratory module using Pioreactor technology (~$400/unit vs. > $10,000 for traditional systems). Iterative refinement revealed a critical design principle: front-loading hardware calibration before biological experime

Bioprinting of transparent and adhesive corneal patches: Integrating photo-crosslinkable dopamine-conjugated silk fibroin and decellularized cornea matrix for sutureless tissue integration and regeneration.

Corneal injuries, a leading cause of visual impairment, are traditionally addressed through tissue transplantation. However, challenges such as donor shortages, graft rejection, and complications from suturing often limit their effectiveness. Current corneal adhesives frequently fall short in both adhesion strength and biocompatibility. We present an innovative solution: a photocurable hydrogel that integrates dopamine-conjugated methacrylated silk fibroin (d-MSF) with a decellularized corneal matrix (DCM). This hydrogel combines advanced materials to create a bioadhesive system that offers su

Biomimetic Microstructured Scaffold with Release of Re-Modified Teriparatide for Osteoporotic Tendon-to-Bone Regeneration via Balancing Bone Homeostasis.

Osteoporotic tendon-to-bone interface healing is challenging, with a high surgical repair failure rate of up to 68%. Conventional tissue engineering approaches have primarily focused on promoting interface healing by stimulating regeneration in either the tendon or bone. However, these methods often fall short of achieving optimal therapeutic outcomes due to their neglect of balancing bone homeostasis and remodeling the microstructure at the osteoporotic tendon-to-bone interface. Herein, a series of site-specific functional modifications are carried out on teriparatide to develop recombinant h

Chromosome-level genome assembly of an endoparasitoid Cotesia ruficrus.

Cotesia ruficrus is a gregarious larval endoparasitoid of many important agricultural pest moths, such as the fall armyworm, northern armyworm and rice leaffolder. While eight of Cotesia genomes had been reported, high-quality genomic data of C. ruficrus has yet to be performed. This study presents a chromosome-scale genome assembly of C. ruficrus, utilizing both PacBio HiFi-reads and Hi-C reads. The genome size is about 185.3 Mb harboring 10 chromosomes, and 209 unanchored scaffolds with a scaffold N50 length of 15.81 Mb. The anchored rate of sequences to chromosomes is 95.55%. A total of 14,

Benchmarking DNA large language models on quadruplexes.

Large language models (LLMs) in genomics have successfully predicted various functional genomic elements. While their performance is typically evaluated using genomic benchmark datasets, it remains unclear which LLM is best suited for specific downstream tasks, particularly for generating whole-genome annotations. Current LLMs in genomics fall into three main categories: transformer-based models, long convolution-based models, and state-space models (SSMs). In this study, we benchmarked three different types of LLM architectures for generating whole-genome maps of G-quadruplexes (GQ), a type o

Genome Report: Improved chromosome-level genome assembly of the American cockroach, Periplaneta americana.

The American cockroach, Periplaneta americana, is a cosmopolitan insect notorious for thriving among humans undeterred by attempts to eliminate it. The traits that contribute to its ubiquity as an opportunistic pest, such as long lifespan, expansive neurosensory capacity, and nutritional flexibility, also make P. americana an excellent invertebrate model organism with a long history in neuroscience and physiological research. Current genetic resources available for P. americana highlight its large, complex genome and richly diverse transcriptional capabilities, but fall short of producing a co

Advances in in vitro cultivation techniques for comprehensive analysis of human gut microbiome.

The role of gut microbiota in human health and disease is becoming increasingly recognized. Historically, the impact of human gut microbiota on health has been studied using clinical trials and animal models. However, clinical studies often struggle with controlling variables and pinpointing disease-causing factors, while animal models fall short of accurately replicating the human gut environment. Additionally, continuous sample collection for gut microbiota analysis in vivo presents significant ethical and technical challenges. To address these limitations, in vitro fermentation models have

Development of dual-function disulfaneyl oxadiazole bactericides by structurally incorporating β-carboline moieties.

Structural derivatization of known leads using the scaffold hopping strategy is a classic approach in pesticide discovery. However, the modificatory universality of lead skeletons profoundly influences the feasibility of this protocol. We herein report development of disulfaneyl oxadiazole bactericides involving β-carboline moieties. Derivative B18 is filtered out to exhibit excellent in vitro antibacterial properties with half-maximal effective concentrations of 0.47 mg/L, 1.72 mg/L, and 5.97 mg/L against Xanthomonas oryzae pv. oryzae, Xanthomonas oryzae pv. oryzicola, and Xanthomonas citri p

Engineered collagen-coated scaffolds for tendon regeneration: a multifunctional drug delivery approach.

Tendon injuries rank among the most common musculoskeletal disorders, with their incidence rising steadily due to increased sports participation and an aging population. Current surgical treatments often fall short of clinical expectations due to poor biocompatibility, suboptimal mechanical properties, and frequent post-surgical fibrosis. Tissue engineering, however, offers promising alternatives by using biocompatible scaffolds that mimic the native tendon structure. This study aimed to develop an advanced tendon-like bundle replicating the microarchitecture and mechanical strength of native
